簡(jiǎn)介:華中農(nóng)業(yè)大學(xué)博士學(xué)位論文亞洲香菇系統(tǒng)發(fā)育學(xué)地位的重新評(píng)估與遺傳多樣性分析姓名徐學(xué)鋒申請(qǐng)學(xué)位級(jí)別博士專業(yè)微生物學(xué)指導(dǎo)教師林芳燦20050701個(gè)形念種,而譜系V卻與澳洲群體的系統(tǒng)發(fā)育關(guān)系更近,這也剛好與“東半球的三個(gè)形態(tài)學(xué)種相互交配可親和”的結(jié)論吻合表明分子系統(tǒng)學(xué)與傳統(tǒng)形態(tài)分類之間可以相互佐證;亞洲群體中譜系V的特殊性預(yù)示著可以將其另立為一個(gè)新種,是否可行尚待深入研究5多樣性中心的確立在全球LENTINULA屬的7個(gè)或者說(shuō)8個(gè)譜系中,中國(guó)既擁有菌株數(shù)量眾多的島嶼一大陸型譜系I,還擁有惟一的大陸型譜系V,而且譜系I還有進(jìn)一步分化成兩個(gè)譜系的趨勢(shì)無(wú)論是從譜系的數(shù)量,譜系的類型還是菌株的分布范圍上考慮,中國(guó)或者說(shuō)是亞洲都是LENTINULA屆重要的遺傳多樣性中心,對(duì)形態(tài)學(xué)種厶EDODES而言,就更是如此6。分子鐘檢驗(yàn)和分歧時(shí)間的估計(jì)從7個(gè)或者說(shuō)8個(gè)不同的譜系中分別抽取部分代表菌株,以金錢菌為外類群構(gòu)建系統(tǒng)樹,以似然比法檢驗(yàn)分子鐘,發(fā)現(xiàn)11RS不具各進(jìn)化速率一致性,拒絕分子鐘假說(shuō)但是仍舊以它估計(jì)支長(zhǎng),結(jié)合前人的研究結(jié)果校正分歧時(shí)間,發(fā)現(xiàn)亞洲主流譜系L與其它譜系的分歧節(jié)點(diǎn)D發(fā)生在420萬(wàn)年左右;譜系1分化成H和IB的時(shí)間大約在39萬(wàn)年左右;以美洲與歐亞大陸分離的時(shí)間和新西蘭與澳洲分離的時(shí)間作為校正點(diǎn),粗略估計(jì)幾個(gè)重要節(jié)點(diǎn)的分歧時(shí)間有較大差異群體遺傳分析1以亞洲群體為主的東半球LENTINULA屬不同聚群的劃分及多樣性區(qū)域的確立T由于亞洲菌株都界定為形態(tài)學(xué)種LENTINULAEDODES,以前述系統(tǒng)發(fā)育分析方法構(gòu)建亞洲范圍內(nèi)的系統(tǒng)發(fā)育關(guān)系,將亞洲群體確立為3個(gè)不同的譜系,根據(jù)不同地域菌株在不同譜系中的分布可以將亞洲菌株分成東北亞、西北高原、西南高原、東部沿海和泛中南等5個(gè)群體結(jié)合巴新和澳洲及新西蘭的2個(gè)群體進(jìn)行綜合分析發(fā)現(xiàn),‘中國(guó)西北高原、西南高原、東部沿海和巴一澳等4個(gè)群體涵蓋的譜系數(shù)多,遺傳多樣性相對(duì)更為豐富,其中中國(guó)西南最為突出2。群體遺傳格局對(duì)5個(gè)亞洲群體和2個(gè)澳洲群體進(jìn)行綜合分析,發(fā)現(xiàn)中國(guó)西南群體的核苷酸多樣度最高,其次為巴一澳,西北和泛中南等3個(gè)群體,東部沿海群體最低群體問(wèn)的關(guān)系顯示T西南群體與東北、東部沿海、泛中南和西北等4個(gè)群體的遺傳分化相當(dāng),遺傳距離接近;西北群體與西南群體的關(guān)系相對(duì)密切,與巴一澳和新西蘭群體的關(guān)系緊密程度較次于西南,而與東北、東部沿海和泛中南的關(guān)系則相對(duì)更遠(yuǎn)一些;巴一澳和新西蘭群體與東北,東部沿海和泛中南等群體的關(guān)系最遠(yuǎn);需要關(guān)注的是巴一澳和新西蘭群體與中國(guó)西北群體的關(guān)系相對(duì)更為密切,但地緣關(guān)系卻很遠(yuǎn)3演化推斷在前述的遺傳格局下,由于巴一澳和新西蘭群體與中國(guó)西北群體的關(guān)系密切,但地緣關(guān)系卻很遠(yuǎn),結(jié)合系統(tǒng)發(fā)育分析結(jié)果研究發(fā)現(xiàn),東半5

簡(jiǎn)介:中山大學(xué)博士學(xué)位論文斜帶石斑魚早期生活史及種苗繁育技術(shù)研究姓名張海發(fā)申請(qǐng)學(xué)位級(jí)別博士專業(yè)海洋生物學(xué)指導(dǎo)教師林浩然20050609卵所含的蛋白和脂肪量自然就高于秋季產(chǎn)的卵。在育苗實(shí)踐中,要充分利用春季產(chǎn)卵質(zhì)量好的優(yōu)勢(shì),大批量開展育苗生產(chǎn)。詳細(xì)描述從受精卵到初孵仔魚的28個(gè)具體發(fā)育時(shí)期的形態(tài)特征和發(fā)育時(shí)間,將斜帶石斑魚胚胎發(fā)育劃分為卵裂期、囊胚期、原腸胚期、神經(jīng)胚期和器官形成期。在水溫2505℃、鹽度310、28H30MIN完成整個(gè)胚胎發(fā)育孵化出膜。PH78的海水中,斜帶石斑魚胚胎歷時(shí)胚后發(fā)育主要根據(jù)卵黃囊、腹鰭棘及第二背鰭棘、鱗片、體色的變化分為仔魚期、稚魚期、幼魚期。仔魚期根據(jù)卵黃囊的有無(wú)又分為早期仔魚和晚期仔魚。在水溫245292℃,海水鹽度288335,PH7585的海水中,培育至36D,發(fā)育最快的斜帶石斑魚結(jié)束仔魚期進(jìn)入稚魚期;培育至42D,發(fā)育最快的稚魚完成變態(tài),成為幼魚。斜帶石斑魚胚后發(fā)育過(guò)程中最明顯的變化是腹鰭棘與第二背鰭棘以及鰭棘上小刺的長(zhǎng)出與收回。在培育水溫2505。C、鹽度3L一32條件下,斜帶石斑魚仔魚孵化后第3D開口并進(jìn)入攝食期,孵化后55D,不能建立外源性營(yíng)養(yǎng)的仔魚分別進(jìn)入不可逆點(diǎn)PNR期,幾乎與此同時(shí),卵黃囊和油球耗盡,仔魚混合營(yíng)養(yǎng)期為2~3D,從初次攝食到進(jìn)入PNR期的時(shí)間為25D仔魚在不同饑餓階段初次攝食發(fā)生率的變化式型是開始較低,此后逐步上升,高峰期出現(xiàn)在卵黃囊接近耗盡時(shí),此后迅速下降記錄到的最高初次攝食發(fā)生率為767%;饑餓仔魚的死亡高峰出現(xiàn)在孵化后46D,至第8D,饑餓仔魚全部死亡卵黃囊期仔魚的生長(zhǎng)可以分為3個(gè)期相初孵時(shí)的快速生長(zhǎng)期,卵黃囊消失前后的慢速生長(zhǎng)期,以及在不能建立外源性攝食后的負(fù)生長(zhǎng)期。在仔魚培育階段每天定時(shí)對(duì)仔魚的攝食作跟蹤調(diào)查,以了解仔魚對(duì)輪蟲、豐年蟲、蒙古裸腹涵的攝食強(qiáng)度和攝食發(fā)生率的變化。結(jié)果表明,輪蟲和蒙古裸腹潘是斜帶石斑魚仔魚培育階段非常重要的餌料種類,前者在仔魚培育前中期占有極其重要的作用,后者在仔魚培育中后期占有極其重要的作用;豐年蟲由于存在營(yíng)養(yǎng)缺陷而屬于仔魚培育階段一種爭(zhēng)議較大的餌料,只能輔助使用育苗實(shí)踐中,在H18之前保證輪蟲的充足供應(yīng)以及在H1927期間維持輪蟲的適量供應(yīng)是必須的,否則很可能導(dǎo)致育苗的失敗在H1518期間保證蒙古裸腹潘的適量供應(yīng)以及在H1926期間維持蒙古裸腹潘的充足供應(yīng)是非常重要的,或者有其它大型枝角類、橈足類取代蒙古裸腹潘也是可以的。II

簡(jiǎn)介:中山大學(xué)博士學(xué)位論文人工養(yǎng)殖史氏鱘繁殖內(nèi)分泌及生殖調(diào)控的研究姓名胡紅霞申請(qǐng)學(xué)位級(jí)別博士專業(yè)生理學(xué)指導(dǎo)教師林浩然劉曉春20060501GTH1與RSII的D亞基和ⅢIII與LH的B亞基構(gòu)成兩個(gè)明顯的聚類。用MEGA2軟件計(jì)算了所比對(duì)物種RHJ外放讀碼框的核苷酸遺傳距離并構(gòu)建了NJ樹和MP樹,陰個(gè)基因0弧基的NIJ系統(tǒng)樹和MP系統(tǒng)樹拓?fù)浣Y(jié)構(gòu)相似,鱘魚與真骨魚類分別聚類,共同形成硬骨魚綱的聚類,’J傳統(tǒng)分類學(xué)一致。成熟肽相似性、遺傳距離和系統(tǒng)樹分支長(zhǎng)度顯示7111IB弧基在真骨魚類進(jìn)化速率皿著快于其他脊椎動(dòng)物,而LJL0亞基在羊膜動(dòng)物中進(jìn)化比其他脊椎動(dòng)物快。暗示這兩種糖蛋白基因的進(jìn)化伴隨物種進(jìn)化而顯示其功能。PCR結(jié)合SOURHEMBLOT結(jié)果表明GTHI和GTILIL0亞基MRNA有廣泛的組織表達(dá),在腦區(qū)的表達(dá)量普遍高于外周組織。G1H【口在脊髓中沒(méi)有檢測(cè)到表達(dá),在腦垂體表達(dá)量最高外周組織中精巢表達(dá)量最高。GTHIIB在腦區(qū)都有表達(dá),仍然是垂體表達(dá)量最高。性腺表達(dá)量較其他外周組織高。NORTHERNBLOT顯示GTHI在腦垂體中有兩個(gè)轉(zhuǎn)錄本,大小約為1167BP和855BP。選用原核表達(dá)拔體PER一22B,分別插入史氏鱘GTIIIIIBⅡ基成熟肽EDNA序列,構(gòu)建成C端含柯6個(gè)組氨酸6一IJS融合蛋白標(biāo)簽的表達(dá)質(zhì)粒;分別轉(zhuǎn)化大腸桿菌表達(dá)菌株B121DE3井誘導(dǎo)表達(dá)兩個(gè)基因。SDS一聚丙烯酰胺凝膠電泳顯示重組融合蛋白分子量分別為GTH【BⅡ基大約14KD,GTHRLB亞基15KD左右。分別用抗組胺酸融合標(biāo)簽的單克隆抗體及兔抗鱘魚71、多克隆抗體對(duì)兩個(gè)表達(dá)蛋白進(jìn)行WESTERNB101分析,結(jié)果顯示目的蛋白分子量與預(yù)期的致,說(shuō)明暈組蛋白有較高的免疫活性。兩個(gè)重組蛋白都是在2小時(shí)就有明顯的表達(dá),6小時(shí)后隨著時(shí)M增加表達(dá)量小再增加;25。C誘導(dǎo)重組蛋白產(chǎn)量比37℃誘導(dǎo)產(chǎn)量低;GTHIB表達(dá)蛋白在上清和沉淀中都存在,可溶性蛋白比包涵體略少大多數(shù)6T1110重組蛋廣1以包涵體形式存在。用蛋白質(zhì)快速液LJJ析FPLC分離鱘魚腦垂體GTII,共獲得;TH純化蛋白4,6MG。反相高效液相色潛RPHPLC分離出兩個(gè)亞基,質(zhì)譜測(cè)定A亞基分子量15603KD,兩個(gè)B亞基分子量分別為14338KD和14694KD。制備了鱘魚汀H的多克隆抗體,并』J“5I標(biāo)記抗原,建立放射免疫測(cè)定方法。測(cè)定了催產(chǎn)前后不同時(shí)間史氏鱘血清中GTH含量的變化,統(tǒng)計(jì)分析表明成功排卵的魚比未排卵和陰性時(shí)照組在催產(chǎn)后12小時(shí)和16小時(shí)GTH分泌量升高顯著。通過(guò)測(cè)定34齡和56齡史氏鱘1卜KT、E2和汀H的周年變化,分析了這幾
