2、to hair follicle growth, in different hair follicle developmental stages, and how this gane affect regulate mechanism of clothing hair, by employing the method of molecular biology techniques and
3、 histology, to do expriment to the skin samples of Qinghai Tibetan sheep living skin. And conclutions from the research results are as below: First of all, by employing the method of RT- PCR
4、, this study successfully cloned cDNA sequence, which is in a length of 381bp, of Qinghai Tibetan Sheep KAP6.1 gene, including a complete CDS area of 252bp and encoding 83 amino acids; and c
5、DNA sequence, which is in a partial CDS area of 356bp, of BMP2 gene, encoding 118 amino acids. The sequence alignment revealed that those two genes have a high degree of homology with speci
6、es like ordinary sheep, goats, cattle, horses, pigs, human beings, mice and so on, and even 100% with ordinary sheep. Second of all, by employing the method of real- time fluorescently quantit
7、ative- PCR, results from the relative quantity analysis of five gene mRNA of VEGF, HGF, IGF- I, KAP6.1 and BMP2 expressions showed that, all those five genes exhibited different levels of expre
8、ssion in skin hair follicles within the experiment year. Among them, IGF- , VEGF and HGF gene were lowest in ⅠFebruary and highest in August; BMP2 gene was highest in February and lowest in
9、 August; and HGF gene exhibited a gradual upward trend from February to November. And at last, by making HE staining morphological ovservation of Qinghai Tibetan Sheep skin, the results indicat
10、ed that, Qinghai Tibetan Sheep skin and hair follicle are in static period in February, rising period in May, prosperous period in August, and gradual decreasing period in November. In the cy
11、cle changes of hair follicle, the change of primary hair follicle density was more stable than the secondary, and the S / P values were obviously lower than other strains of sheep. Besides,
